The Serbian says he is ready for the "biggest challenge one can have" at Roland Garros, with Nadal dubbed the "King of Clay".
Tuesday's contest is due to start at 19:45 BST, and both men will be highly confident ahead of their record 59th meeting.
Nadal has not played Djokovic since he lost in last year's semi-final in Paris, and the Spaniard trails 30-28 in the head-to-head record.
The winner will not have to play world number two, Daniil Medvedev, if they advance after he suffered a surprise 6-2 6-3 6-2 defeat to Croatian Marin Cilic.
Fourth seed Stefanos Tsitsipas is also out after he lost 7-5 3-6 6-3 6-4 to teenager Holger Rune.

In the women's draw, top seed Iga Swiatek advanced as she beat China's Zheng Qinwen.
The 21-year-old dropped her first set in ten matches but recovered to record a 6-7 (5-7) 6-0 6-2 win.
Wednesday's first match will see Italy's Martina Trevisan face Leylah Fernandez of Canada, while the other quarter-final is an all-American clash between Coco Gauff and Sloane Stephens.
