Peniston beat Swiss world number 95 Henri Laaksonen in his Wimbledon debut on Monday and he's set to take on Steve Johnson on Wednesday.
The British number six, who beat French Open finalist Casper Ruud during a fine run to the quarter-finals at Queen's last week, is due on court at 11:00.
Peniston is the first of six Brits in action on the third day of the 135th edition of Wimbledon, with Norrie, Murray, Dart, Raducanu and Watson also looking to advance.
Norrie (66/1 Wimbledon outright) takes on Spain's Munar at 16:00 with Murray (33/1) set to square off against big-hitting American Isner at 17:00.

Dart takes on Masarova in her first round match after their initial encounter was postponed, while Raducanu (16/1 Wimbledon outright) and Watson face tough tests against Garcia and Wang respectively.
Away from the British interest, defending men's champion Novak Djokovic steps out against Australian Thanasi Kokkinakis at 13:30.
Djokovic (8/13 Wimbledon outright) required four sets to get beyond South Korea's Soonwoo Kwon in the opening round but he defeated Kokkinakis in straight sets in their only previous meeting.
Carlos Alcaraz (14/1 Wimbledon outright) is another potential contender for the crown at SW19 and he takes on Dutchman Tallon Griekspoor at 16:00.
